Overview
MOTS-c is one of the first recognized mitochondrial-derived peptides — a 16-amino-acid signaling peptide encoded in the mitochondrial 12S rRNA gene. Unlike nuclear-genome-encoded peptides, MOTS-c originates inside the mitochondrion itself, acting as a retrograde signal from mitochondria to nucleus. Why mitochondrial-origin matters: MOTS-c activates AMPK, upregulates PGC-1α, and enhances insulin sensitivity through pathways that parallel the cellular response to exercise. It’s been called an exercise mimetic in the metabolic-research literature. IF the research endpoint is mitochondrial function, insulin sensitivity, or exercise-pathway pharmacology, THEN MOTS-c is the mitochondrial peptide with the densest published evidence.
