Overview
PT-141 (bremelanotide) is the MC4R-selective fragment derived from Melanotan-II. It’s the only FDA-approved libido-targeted pharmaceutical that acts on the central nervous system — specifically on hypothalamic melanocortin-4 receptors — rather than on vascular smooth muscle like the PDE5-inhibitor class. Why the mechanism matters: PDE5 inhibitors (sildenafil, tadalafil) enable erection mechanically by relaxing corpus cavernosum smooth muscle. PT-141 acts upstream on the desire/arousal signal itself. Researchers studying HSDD (hypoactive sexual desire disorder) need a tool that isolates the central mechanism.
